Innate Immune Circuits in Skin Disease — From Target Discovery to Early Clinical Translation

The skin is a particularly useful paradigm organ for elucidating core principles of immunity and immunopathogenesis and for developing novel therapeutic strategies. I will present the research architecture established at the University of Lübeck to dissect pemphigoid diseases as models of autoantibody-driven, granulocyte-mediated inflammation, and illustrate a successful translation: identification of leukotriene B4 as a key pathogenic mediator and its advancement through clinical development to a Phase III trial. Looking forward, I will outline a research program that cross-references different types of skin inflammation and translates these insights into precision therapies. My proposed five-year plan will embed multi-omic cohorts, harmonized SOPs, and biomarker-led Phase Ib/IIa studies across CHUM and CHU Sainte-Justine. Expected deliverables include validated biomarkers, interoperable biobanks, and pilot trials ready for scale-up, and partnerships with local stakeholders planned.
